So I’m looking into building a custom cube. I’ve only built one aquarium before (4.8 gallon whiskey box betta tank) but the techniques there aren’t really applicable. Anyway Amazon has these 24x24 half inch thick plexiglass plates cheap. I’m thinking three pieces of plexiglass, two plywood and some wood strips.
Red stripe and solid red is a 24x24 half inch thick ply
Teal is the 24x24 half inch plexiglass
And yellow is .5x.5x24 hardwood oak maybe.
My real question is what’s the best way to seal and glue/“weld” the plexiglass to the various woods?

Will this be a display tank?

As far as constructing it, you could route half inch slots into the oak, then lock it in with a silicone. All wood will need sealed (it could leach and absorb water.) Other than epoxy ( and probably several layers) I don't know what you'd seal it with.

Not that I've ever done this. But you do have my curiosity piqued.

Definitely my display. If my measurements are correct it’s going to be roughly 55 gallons plus I’m contemplating using my original 13.5 gallon display as a sump. I’d have to make a custom stand but I do have some carpentry experience.
Oh and here’s a picture of the whiskey tank . Several things I’d do different next time but not bad for a first… well second diy build (my original adhesive didn’t hold and the plexiglass blew off)

Right on. What if you switched the corner posts from 1/2 inch to say 2" x 2" then route a rabbet into the sides holding the panels. Then it's edges will be captured on 3 sides. Plus the volume/mass of 55 gallons is quite a bit. It may help with some sturdier materials.

Plus a list of adhesives for various materials. I can't vouch for there ability to withstand constant contact with saltwater. But it could give you a start on what to look for.
