If you put live sand into straight RO you possibly killed most of the beneficial bacteria that was in the sand. Also if the Rock becomes water logged with non salt water it may take a bit for the bacteria to colonize the center pores as well. Get some salt water in it, Auto Top off set up and start ghost feeding. I personally am a fan of mixing Dr. Tim's and Fritz turbo start to get the good bacteria. As has already been said Patience in this hobby is key.

Oh man I didn't realize you haven't added salt yet!
Lack of patience will put you out of this hobby in short work.
It's going to be very hard to get any salt you put in to mix correctly with sand and rock already in the tank. You've definitely set yourself back a bit.
Was that RODI water you put in or tap water?
