Bean Animal Plumbing Question

I have a 8' long 240 gallon I am about to start plumbing. The tank had a external overflow box setup for a bean animal system with 1-1/2 bulkheads. I am currently planning on a Triton recommended setup so 10x flow through the sump and refugium. Return will most likely be dual Varios 8's plumbed into Sea Swirls on the back 2 corners. Tank stand is 34" tall to the base of the stand.

I have a fish room for this build but it is to the side of the tank. So plumbing will need to come out of the overflow box and run around 6' to the side through a wall to the fish room. I remember it being said the siphon line needs to have some drop to it to help purge the air. What do y'all think of a short vertical drop then 2 45 degree elbows transitioning it to horizontal at the height I need the drain line for the sump? Or would just a single 90 degree elbow be fine?
 
Avoid running horizontally. Is there a way you can use a single 45 at the beginning and have it run downhill far enough into the fish room?
