My wrasse has had this brown spot near his anus for a few days now. Is he just constipated or is it something else?

Are there any other symptoms to go along with the spot? If it's intestinal worms, then you will see white stringy poop. Other symptoms to look for are: lethargy, lack of appetite, breathing heavily, flashing or scratching, yawning, head shaking or anything else very weird in appearance and behavior.

Nope, no other symptoms and I haven’t seen poop of any color. He just finished a 30 day treatment of copper safe about a week ago. Not sure if that would be related.

I'd be more inclined to think it's an infection or internal hematoma (my spelling stinks). For now, if you aren't already doing this, be sure to add vitamins to his/her food and keep a close eye on it. If it gets bigger, worse, or the wrasse start displaying any other symptoms like I listed then be ready with antibiotics like kanaplex or furan2.

The fish is still acting fine but the spot doesn’t look any better. I think I will go ahead and treat. Do I give kanaplex in the water or in his food? Should I give him some metroplex also?

Kanaplex would be dosed in the water of the QT. Don't use the metroplex until it's needed. If you see white stringy poop, then you'll feed it in the food. If the kanaplex alone doesn't help after several days, then you can add the metro to the kanaplex.
