Type of silicone is very important. I built my own 20 gallon cube and after a bunch of research on silicone I ended up with RTV 100 series from grainger. Has a super strong bond and is aquarium safe.

I would imagine it would as long as you use the right size lumber. 2x4s probably wouldn't cut it on this size.

Building your own reef tank is fun. Doing anything for the first time can be difficult depending on your own skill sets and patience.
Timber is an exceptional material to build a support structure for a small aquarium tank like this one. In my view often better than steel as it does not corrode and can be easily modified.
Timber selection is paramount as is the design. Without you posting your design concept, hard to say for sure, go for it.
But a rough rule of thumb for you.... 2x4s are fine based on a 2' span on a tank of this depth. Providing you do not envisage a heavy lighting shroud or heavy over tank cabinets...?
Aged structural pine will be more than adequate. Cross tie the legs to prevent sway. This can be done with permanently attached exterior marine ply panels or cross ties. Then treat and water proof the timber with your chosen finish.
