I tried that last night. I'll do it again now
I don’t know much about that particular pump but if the lines are clear and the rollers are rotating a peristaltic pump will suck water from a long distance. They do not need priming. If the water isn’t flowing with the bucket test and the pump appears to be operating correctly, I would take the tubing out of the pump head and make sure it’s not totally flattened. Then reassemble and test again.

Looks like that gizmo with the white piece puts pressure on the tubing when it’s in use position. You mentioned putting water into the pump with your mouth. Are you able to suck water through or blow air through with the pump off? You shouldn’t be.
 
There is some voodoo in getting that reactor to work correctly - especially if the reactor is above your sump.

What works best for me is to use a powerhead to push water to the reactor. Then.... use the kameor to control the flow as it leaves the reactor. While it is in the "pulling position" - it's acting more like a controlled resistor - it does not work well if you truly try to pull the water and run the whole thing at negative pressure - in my experience. You will end up with air intrusion.

When you push... your reactor pressure will be limited by the pump head pressure. So, no worry with a propeller pump, but, you have to be a little more careful with a peristaltic pump or pressure rated pump in the pushing position.

So... sump -powerhead - reactor - kameor - sump

Did you try screwing the screw tighter. It might be to loose.
That’s what I was wondering. If there isn’t enough pressure squeezing the tubing as the rollers spin the pump won’t pump. Is that little screw knob just to right of that white piece adjustable?
