i have a Neptune dos and was wanting to do auto water changes. I have a 180 gallon system how many ml a day would be recommended

Kinda unclear..
You want to automate water changes on a 180, it would come out to how many gallons a day imo.
Automating water changes is tricky, easy to get out of tank balance.
Or are you asking automated top off?

So the ones who use the dos system do they do it all in one shot or do they do it multiple times a day

Mine runs slowly throughout the day. I'm exchanging 45oo mL per day, and it breaks this up automatically into 118 doses of 38.1 mL per day. I'm struggling with this. My first issue is, the pumps are recommended to be re-calibrated every 100 hours of use, which isn't that long when using it as a AWC system. I've re-calibrated twice and there have only been variances of 0.5 mL. Also, since it's replacing water so slowly throughout the day, there is a certain percentage of water being removed that was just put into the tank earlier, meaning you'll actually need to exchange a larger amount of water to achieve the preferred results even if your input and output are placed well apart from each other.

So how long would it take to take out like 4500 ml then replace 4500 ml? Would that be more beneficial?

I suppose you could in conjunction with turning off equipment such as your skimmer and ATO. That is always an option, but the DOS has the capability to run one pump in reverse to simultaneously do it, which is still, with regards to my last comment, my preferred method. As to how long it will take, the pump is capable at running at different speeds indicated by a different mL/min ratio. In all actuality, you could remove 4500 mL in just a few minutes, then replace it a few minutes later, as long as your pumps remain calibrated and your NSW matches salinity of your OSW. Choose your journey.
