Help with DOS water change

I have a very important question about setting up my auto water change system.
In the diagram you can see that the DOS will be 88feet away from the tank, the question I have is where to calibrate it from. Should I make the long runs with my ro/di line and then calibrate or do it before I run any line?
I need to make sure I am taking out the same I am putting in. doslayout.jpg
My tank is an 80g deep blue so I am figuring probably one big change of 25% cause it has just finished the 2nd cycle and then 12g a week after that.
 
My setup is a bit different... sump, NSW reservoir, and DOS are no more than a few feet apart, but my drain line runs nearly 100 feet.

Believe it or not... calibrating the dos with, and without the 100 feet of RO/DI line on the drain side makes zero difference.

Wow that is amazing, but it seems after you prime the line whether it is 6in or 80 feet it will pull the same amount regardless.
