Help with quarantining?

I'm starting up a 29g barebottom fowlr, I currently have ~30lbs LR curing. There's a hob filter running with carbon (just in case) and floss. I plan on using an extra 10g as a QT tank, and the hob with the filter floss to stave off a mini-cycle when I introduce my first fish.

I plan on adding a royal gramma first, then a percula clown, then one more small fish I haven't yet decided on (feel free to make suggestions!). My question is, does the gramma need to be QT'd in the 10g before I add him to the DT, since he's first?
 
Yes! If he has ich or brooke it will infect the new tank. Any future fish placed in the DT will be at risk of contracting it. The solution to such a scenario would be to catch all the fish in the DT and QT/ treat and allow the DT to remain fallow for 72 days. Problem is you can never be sure if the fish has it to begin with. A healthy fish may carry enough of the parasites to infect your tank but never show signs of infection. I actually made that mistake when I bought my first clown fish. I figured no other fish in the tank so should be no big deal right? Wrong!
