Does the gate valve on the main overflow have to be installed on a vertical piece of pipe or could it be installed on a horizontal piece instead?

It can be horizontal, vertical or diagonal. Whatever you like. Just serves to restrict flow so there is a solid column of water proximal to it.

You should limit the length of horizontal run on a drain pipe, which also would reduce the number of 90 elbows you use

Horizontal is fine. One of my tanks has the valve in horizontal section.
