You need to wait long enough for it to evenly disperse, like food coloring becoming all one color/intensity.
In some tanks that might be a few minutes, and in some with extended sumps and refugia that might mean hours. It will also take a much longer time to get into pores in rock and sand, but that is unlikely to make a big difference in many tanks.
How far off was it? What did you add?
