I believe this is either mulm or the star of chrysophytes.
Try to dislodge with a turkey baster and siphon up the film matter. if it doesnt dislodge, you will have to remove rock and place in a bucket of tank water and scrub with a toothbrush (not as labor bad as it sounds)
