I just set up my Waterbox 100.3 and there seems to be a leak in the return line. All the O rings and rubber gaskets seem to be in place and I think I have it pretty tight, what else could the issue be? And if I don't get it fixed, theoretically is it fine to just add something so that the leak flows back into the sump instead of onto my floor behind the aquarium?

Do you know which connection it's leaking from? Be aware that overtightening can cause leaks as well. I wouldn't start a tank up with a leak though, asking for trouble no matter how tempting it is.
