!!! Welcome to R2R @nagili !!! When you said a reef your meaning a ocean reef or another reefer tank ? I don't think you will need to wait too long but I would get a Seachem Ammonia Monitor Batch and keep eyes for ammonia level until reading get 0.

Yes Live rock only becomes live after the rock has established that bacteria layer and saltwater may contain some small amount of bacteria but it is not very much at all. Like @spyder813 stated check your ammonia levels as well as Nitrite and Nitrate and you will know when the cycle is completed. There is a ton of information on this site about the aquarium cycle.

I mean from an ocean reef, here in Maldives it is easy to get live stocks from sea, I want build a 36”x24x24 reef tank with Corel and fish, I can get those from sea, that’s why asked. Actually I don’t have the chemical knowledge

Ok, in that case you just need to check for any unwanted hitchhiker that may comes with that rock. That's almost instant cycle, especially if goes straight from the ocean in a bucket to your tank. I have a friend in Australia that did the same and didn't have any issue.

With that rock and water the only thing to do is change out 10% of the water once a week. Dont add too many fish at once.

In design sump part is enough for the measurement? I plan to fit 2 filter socks in 2nd and 3rd chamber, 1st part is the intake from tank, 4th is refugium and last part is return

It would help if you labeled the chambers 1-5. My sump doesn't have chambers, but if you have a refugium it is good to separate that from the pump. Fewer chambers allows for more ability to move equipment later if you want to?

Here in Maldives normal temperature is 30c+ daytime, if I use a copper coil in the tank to chill the saltwater is it harmful or which material I should use for this purpose, I want build chiller using a small refrigerator
Copper is not good. Salt water is very corrosive to metals. Stainless steel would be best if you need to pump salt water thru metal. It is very hard to bend. Another option would be to use plastic hose.
