Need help with starting MicroBacter Lettice

I have a ~100 gallon system and I have very high nitrates 100+ ppm i have tried feeding less changing my flow rock work add biodigest nitrifying bacteria with no help. I thought maybe i need a hand with anaerobic bacteria surfacing. I paid a visit to my local store and found the Microbacter by brightwell aquatics added to a filter box as seen below and rinsed with aquarium water afterwards I added two vials of Prodibio biodigest to the filter box and kept it as is thinking of maybe the bacteria might reproduce. Im thinking of letting water circulate a few drops per second to reduce oxygen entrance to the filterbox and help anaerobic bacteria growth. I have been adding NoPox by redsea and couldnt control nitrates now I have dinoflagelletas all over the aquarium. Im going crazy but dont know what to do with all limited resources here in Northern Iraq.

So the question is shoukd I let the filterbox run as a drop system or should I let water run and how should I place it i mean where in sump.
