I have a 600 gallon, wife purchased a lil 10 gallon nano cube. She took water and rock from my 600g display and a setup her 10g nano. Does it need to cycle or should it already be cycled? Also, she added new sand since my 600g display is bare bottom

Add some ammonia and test. That should tell you if it's gone tomorrow.
 
I'd try 4 to 8 drops and test. 4ppm would be a good starting point. I think that's the level Dr Tim's uses to start with so aim for that.
 
I did the same thing for my 20 gallon coral QT and added a bottle of Bio Spira just to be safe. It has been up and running for a little over a month now with no issues at all. Never saw any signs of Ammonia or Nitrite at all, (I tested every day for the first couple of weeks to be safe).
