Removing Tank Off System?

I have a 120g tank with a 40g frag tank plumbed together. Now that I work remotely full-time I've been thinking about having a larger tank in my office with me, and the 40g frag would work well. My only question is whether or not this is going to make things go haywire?

Tank has been running for nearly 2 years and they've been plumbed together for the full duration. I think total system water volume is approximately 160g including the sump.
 
if they remain plumbed together, i do not see an issue with it long term. however, the move itself will cause some setbacks in regards to tearing things down, moving them, and then reconstructing them.

My plan would be to separate them. I have a 20g I can use as a sump.
