Return line up and down?

No big reason not to. Aside from aethetics it wont be much different than strait up into the tank.
 
Actually thats a myth. Brs did a video showing elbows dont reduce flow significantly at all.
 
Up snd down is the way I know it when it comes to return lines especially for maximum flow
 
Testing friction loss is one thing and I agree it's probably negligible for most installs; however, for a long term setup I would want as few fittings as possible to prevent crevices that allow growth of organisims.
