I am setting two of them up that way on my 125 build. I just ordered them and the adapters last week. No water in the tank yet though. 
If it doesn't work (which I don't see why it wouldn't) then it is easy enough to take the nozzles off.

3/4". I will be running a separate pump on each one, for redundancy (should one go out while I am not around). I am using a couple of Vectra M-1 pumps. They will each be running at less than capacity and that may inhibit the performance of the RFG nozzles, but we will see.

That’s kind of what I had in mind, but I’m thinking of just running 1 swirl. The tank is an old Aqueon with the dual overflows in the back. I’m gonna use the overflows for drains then run returns over the back. Thought I’d place the swirl in the middle back of the tank between the overflows and use one pump to feed it and a manifold. The other pump would feed 2 regular returns feeding the 2 back corners. I am also planning to purchase 2 gyres for the sides.

It isn't "if" they will work, they will. It is more of a question on how you elect to mount them. If you do a search you will see people are using them. I've been interested in them with my 1" sea swirls but not a fan of the additional length they add using the default elbow the SS ships with. I know VCA is working on a better solution to address this - several have raise their concern. Should be out soon if not already - can always send them a email to ask for status.
I love my sea swirls but the VCA nozzle isn't has urgent at the moment since I have a pair of XF250 gyres. If I find a creative way to mount them while addressing my concern on the added length of the nozzle I'd probably go that route and sell the XF's.

I'm setting up an Aqueon 210 as well and plan to do dual herbies with the stock overflows and run my return up and over. That got me to thinking about sea swirls and I use VCA nozzles on all my tanks so I'll definitely be giving that a try. I was only planning on doing a single return with a pair of gyre xf250's to provide the movement inside the tank. Maybe put it in the center of the tank and use a y adapter to get a full 180 degree coverage.
I've never used a sea swirl before though so I'm not sure if that will work.

Sorry for being so late to this thread - just happened upon it. We have a number of users that have added the RFG to the end o the Sea Swirl with good success.
The best way to add the RFG is with a Loc-line NPT fitting threaded directly into the elbow that comes with the Sea Swirl. For the 1in Sea Swirl, the only option we had used to be a 1in Slip-Fit Nozzle, which does had a bit of length to the stock Sea Swirl.
However, @saf1 is correct, in that we had been working to create a better solution. The result is the new RFGS100LL We where able to source out a true 1in NPT fitting which threads perfectly into the 1in Sea Swirl. The new RFGS100LL is shorter than a standard 1in RFG. In fact it's about the same length as the 3/4in RFG allowing it to take up much less room.

yep - it's already available. For the 3/4in you just need a 3/4in RFG nozzle, and then pair that with the 3/4in NPT fitting . We don't package them together however.
I've even seen where one user tool their 1in Sea Swirls and added one of those 90 3-way splitters and added dual RFGs to the end.
Perfect and thanks. Will check the dimensions when I get home because I don't want it to be longer than the setup I have now.
Here's a few shots of the the 3/4in RFG with an NPT fitting next to the new 1in RFGS with the 1in NPT. As far as adding nozzle to the end of a Sea Swirls goes, it's not too bad. The RFG will only add about 5 inces to the overall length.
