Should I be worried?

So I placed my 40g on top of the stand I built. Although it is empty I’m showing a gap at one of my corners. Will it level out once it’s filled?

A flat piece of glass will bend to fill that gap. This means the glass will be stressed. Can cause it to break or the seams to separate.
 
I'd play it safe and fill it slowly as a test. If it holds water, let it sit for a few days just in case. Your choice because some people won't want to risk it.
Add a leveling pad under the tank. My favorite is yoga mats.

Don’t risk it get a mat there pretty cheap
I’m asking purely because I am new here. Is this something that is a big deal? It seems like the frame corner is holding a very insignificant amount of weight. I’m not questioning just looking for knowledge
 
Odds are it will be just fine. It's only a 40g tank and not really that much of a gap. Not saying anyone should do this, but I have kept numerous 40b breeders as QT tanks on way less level or flat stands than what you have got with never a problem. You are right to notice that the bottom frame of the tank is helping to distribute the load evenly across the bottom. A big gap might be a problem, a little one like that is within the tolerances of the tank.

But to be extra safe, take the advice of the yoga mat, it's a good suggestion.
 
You may never have a problem but a crack in a bottom panel could happen at any time. And empty your whole tank. For the few bucks for a mat it's worth the insurance.
