So I just had my aquarium rebuilt and I filled it with water and sand already and there's a small leak about one drop per two seconds from the base of the Overflow leaking in like it's coming in from under the silicone.

Besides draining the tank again in moving the sand back in reselling it is there anything I can do to stop that leak from the inside of the Overflow?

No.
What other repairs were done on the tank?

The reason I am asking is because if the overflow was the only repair then I personally would not worry about a small leak in the overflow. The only concern would be a prolonged power outage that could possibly overflow your sump, one drop per second would take hours so I would not be overly concerned..

Can you apply silicone to the inside of the overflow?

You could glue your overflow pipes into the bulkheads, creating a seal at the bulkhead/pvc connection inside the overflow box. Then, it doesn’t matter if water leaks into the overflow box. The water will stop draining into the sump when the water level gets below the tops of overflow drain pipes. Does that make sense?
