I am looking for design help on a t-slot aquarium stand for a 300 gallon aquarium. It is 96 x 24 x 30. I assume it will need to hold at least 3500 lbs. What size/kind of t-slot do I need? Will one center brace be enough? How many cross supports on the top and bottom? What fasteners should I use to ensure the joints are strong enough?

I would recommend you contact the t-slot manufacturer and discuss this with their engineering team. Misumi and 8020.net both offer design assistance. When you start looking at high loads such as a tank this large, steel becomes much more attractive as you will likely be able to build a stand with smaller diameter steel tube than you would with aluminum.
I love t-slot for tanks under 150 gallons, but anything over I would go with welded steel.
You should also factor for 4000 lbs to be safe. 300 gallon aquarium:
Glass = 700 lbs
Water = 2600 lbs
Rock = 300 lbs
Sand = 150 lbs
Sump/equipment = 300 lbs
