New 29 biocube has been cycling for 3 weeks with live sand and 25lbs live rock. At what point should I do my first water Chang? What amount? Thank you very much.

Since you are using live rock (( and I am assuming that here is "life" on it )), I would have already done a water change. I, personally, like to limit the ammonia spike so that I preserve much of the life that I paid good money for.

In addition, this way the bacteria growth won't over shoot the carring capicity of your tank.

Yes it has cycled and live rock does have life on it. Can you tell me how many gallons I should change? It has about 20gl in it with displacement.

I would change the amount that is easiest for you to do on a regular basis.

All 3 of my tanks (( 40, 65, 75 )) get 5 gallon water changes, since I have a bunch of 5 gallon buckets laying around. It is a easy size to work with and carry around. IMO/E, anything b/t 2 to 5 gallons should work, just pick the one that is best for you and your tank.

Remember, the easier it is to do a water change, the more apt you are to do it.
