Why won't this drain?

Im attempting to run a bean animal, but drain number 2 will not dry like it's air locked or something. Check out the pic you can see numbers on the drywall. The only thing weird or different from the rest is a slight half inch bend back up Hill before dropping in to the floor but the bend is still below the box....

And I'm not gonna lie its a little sketchy running this tank currently with no leak sensors with my new room and carpet next door. Maybe you can help with some ideas

You are expecting water to defy gravity and run uphill. Regardless of the number of drain lines you have - 1x for Durso, 2x for herbie, 3x for bean animal - they are all gravity drains and must run downhill.
 
I would give those designs more of an initial vertical drop out of the overflow... and then do away with the uphill bend...air is trapped in there and thou don't have enough pressure to push it out...especially without the bigger vertical drops out if the overflow box .
